Aunque a menudo se mencionan juntas, miden aspectos diferentes. Conocer esta distinci\u00f3n facilita la interpretaci\u00f3n de cualquier certificado de an\u00e1lisis.<\/p>\n<h2>Lo que mide la HPLC: la pureza<\/h2>\n<p>La cromatograf\u00eda l\u00edquida de alta resoluci\u00f3n (HLPC) hace pasar una muestra disuelta a trav\u00e9s de una columna de relleno a alta presi\u00f3n. Las diferentes mol\u00e9culas atraviesan la columna a distintas velocidades en funci\u00f3n de la intensidad con la que interact\u00faan con el material de la columna, de modo que el p\u00e9ptido principal se separa de las impurezas relacionadas. Un detector registra cada componente como un pico. Al integrar el \u00e1rea bajo el pico principal en relaci\u00f3n con el total, el analista calcula el porcentaje de pureza.<\/p>\n<p>Por lo tanto, la HPLC es un <strong>pureza<\/strong> herramienta. Te indica qu\u00e9 proporci\u00f3n de la muestra corresponde al compuesto dominante en comparaci\u00f3n con el resto, pero por s\u00ed sola no demuestra cu\u00e1l es realmente ese compuesto dominante.<\/p>\n<h2>Lo que mide la espectrometr\u00eda de masas: la identidad<\/h2>\n<p>La espectrometr\u00eda de masas (MS) ioniza las mol\u00e9culas y mide su relaci\u00f3n masa-carga con gran precisi\u00f3n. En el caso de un p\u00e9ptido, la masa medida se compara con la masa te\u00f3rica derivada de su secuencia de amino\u00e1cidos. Una coincidencia cercana confirma <strong>identidad<\/strong>: la mol\u00e9cula es, efectivamente, la secuencia que figura en la etiqueta, y no un p\u00e9ptido diferente de tama\u00f1o similar.<\/p>\n<p>La espectroscopia de resonancia magn\u00e9tica (RM) es una herramienta de identificaci\u00f3n. Resulta excepcionalmente eficaz para confirmar que \u201cesta es la mol\u00e9cula correcta\u201d, pero no es el m\u00e9todo principal para cuantificar la pureza.<\/p>\n<h2>\u00bfPor qu\u00e9 se utilizan ambos juntos?<\/h2>\n<p>Analicemos las lagunas que deja cada t\u00e9cnica:<\/p>\n<ul>\n<li>La HPLC por s\u00ed sola podr\u00eda mostrar un pico \u00fanico y perfectamente puro\u2026 pero del p\u00e9ptido equivocado.<\/li>\n<li>Solo la espectrometr\u00eda de masas (MS) podr\u00eda confirmar la masa correcta, mientras que la muestra solo tiene una pureza de 80%.<\/li>\n<\/ul>\n<p>Cuando se utilizan conjuntamente, la HPLC responde a la pregunta \u201c\u00bfqu\u00e9 grado de pureza tiene?\u201d y la MS responde a la pregunta \u201c\u00bfes la mol\u00e9cula correcta?\u201d. Un certificado de an\u00e1lisis (COA) que incluya ambas t\u00e9cnicas ofrece una visi\u00f3n mucho m\u00e1s completa que cualquiera de ellas por separado, raz\u00f3n por la cual la combinaci\u00f3n \u201cHPLC + MS\u201d se ha convertido en el est\u00e1ndar pr\u00e1ctico para la documentaci\u00f3n de p\u00e9ptidos de grado de investigaci\u00f3n.<\/p>\n<h2>Comparar los dos resultados uno al lado del otro<\/h2>\n<p>En un informe combinado deber\u00edas ver un cromatograma de HPLC (a ser posible, un pico dominante n\u00edtido con un porcentaje de pureza indicado) y un espectro o una tabla de MS en la que figure la masa observada junto a la masa calculada. Comprueba que la masa observada coincida con el peso molecular que figura en las especificaciones del producto y que el valor de pureza se ajuste al grado que has solicitado.<\/p>\n<h2>Conclusiones pr\u00e1cticas<\/h2>\n<ul>\n<li>Trata la pureza (HPLC) y la identidad (MS) como dos cuestiones distintas.<\/li>\n<li>Se prefieren los lotes documentados mediante ambas t\u00e9cnicas.<\/li>\n<li>Compara la masa observada (MS) con el peso molecular publicado.<\/li>\n<li>Siempre que sea posible, comprueba las cifras cotej\u00e1ndolas con un informe de un laboratorio independiente.<\/li>\n<\/ul>\n<p>Una vez que tengas bien claras las dos t\u00e9cnicas, el certificado de an\u00e1lisis (COA) de un p\u00e9ptido de investigaci\u00f3n deja de ser una sucesi\u00f3n de n\u00fameros y se convierte en una verificaci\u00f3n r\u00e1pida y fiable.<\/p>\n<p><em>Los compuestos mencionados son materiales de referencia de calidad para investigaci\u00f3n, destinados exclusivamente al uso en laboratorio; no son aptos para el consumo humano ni animal.<\/em><\/p>","protected":false},"excerpt":{"rendered":"<p>La HPLC y la espectrometr\u00eda de masas responden a dos preguntas diferentes sobre un p\u00e9ptido.
